Virtualenvwrapper is great but it's more useful on a box that you'll be developing on for a long time instead of something like a vagrantbox that's meant to be thrown away.

That said, if you want to use virtualenvwrapper, go for it. Nothing will be different other than how you activate the virtualenv.
